Like several others I have used TwitterFon exclusively on the iPhone. I was even considering making the upgrade to TwitterFon Pro. Now I have a new reason to reconsider.
I love the sync with the desktop TweetDeck! I love the different columns and searches and groups. Overall it's a great Twitter client for iPhone
3 Things I wish would change...
1) I would like to be able to change the colors like you can on the desktop app
2) I would like to do landscape, especially since all the core iPhone apps are going to that now
3) I would like the ability to pick the user name from a pop up when I want to mention someone in a tweet instead of having to type it out, like you can in TwitterFon

agree with @postalUT, twitterfon has been my favorite so far but I am looking forward to what tweetdeck has to offer with the desktop sync. Though, they need to improve stability. Better with OS3 maybe?

It's come closer than any other app to replacing twitterfon for me. I just think the process of using twitterfon is much more intuitive and useful than any other twitter app. All you have to do is start the app and then it scrolls down to the oldest unread tweet. It even highlights replies. No other app I've used has been able to do this (though if you know of one let me know). I hate opening up the app, scrolling down 4 pages, then the reading can begin. Plus the stock black on cyan theme of twitterfon is easy for me to read outside on my smoke breaks than the dull dingy brown and grey themes of other clients. I do love the desktop synchronization so I don't have to re-read tweets.

Love it! I thought Tweetie would one day take over the world but TweetDeck for the iPhone is incredible. All it needs to be perfect is push notifications and username autocompletion.

It's great and it has the potential to be awesome! it is buggy and crashes but the features are there and managing multiple accounts on a free twitter app is HUGE! You can also view profiles, follow and unfollow people. The ability to sync with the FREE desktop app is also huge! GREAT work and can't wait for the updates to get better.

I still prefer Tweetie, especially for users with only a single account.
